UAS regulatory sequences drive expression of a mutant form of robo1 in which the region between amino acids 891 and 973 has been swapped with the analogous region of fra. The construct has a signal sequence (Tag:SS(wg)) followed by three Tag:HA tags at the N-terminal end and the Tag:V5 epitope and Tag:polyHis tag at the C-terminal end.
fra::robo1SD.UAS.Tag:V5,Tag:polyHis,Tag:HA,Tag:SS(wg)/Scer\GAL4insc-Mz1407 is a suppressor of abnormal neuroanatomy | embryonic stage phenotype of robo11
fra::robo1SD.UAS.Tag:V5,Tag:polyHis,Tag:HA,Tag:SS(wg)/Scer\GAL4insc-Mz1407 is a non-suppressor of symmetrical commissure | ectopic phenotype of robo11
Expression of fra::roboSD.Scer\UAS.T:SV5\V5,T:Zzzz\His6,T:Ivir\HA1,T:SS-wg under the control of Scer\GAL4insc-Mz1407 almost completely rescues the ectopic commissural crossing phenotype seen in homozygous robo1 mutant embryos. Ectopic crossings are only seen in 20% of segments.
When fra::roboSD.Scer\UAS.T:SV5\V5,T:Zzzz\His6,T:Ivir\HA1,T:SS-wg and commScer\UAS.T:Hsap\MYC are co-expressed under the control of Scer\GAL4insc-Mz1407 the axons collapse into the midline.
