    The number of acidic autolysosomes (detected by LysoTracker staining) is strongly reduced and the number of autophagy structures is increased in fat body cells of starved Mon1mut4/Df(2L)ED7853 mutant third instar larvae relative to controls, while the cells show abnormal accumulation of autophagosomes. Similarly, cells in Mon1mut4 mutant somatic clones in larval fat body display fewer acidic autolysosomes and more autophagosomes than the surrounding control cells.

    Cells in homozygous clones in the wing disc contain enlarged endosomes which are strongly acidified. The number of cells in a given area is increased in the mutant disc compared to wild type.

    Somatic clones of fat body cell double mutant for both Rab52 and Mon1mut4 are larger than their neighboring control cells (as are Rab52 single mutant cells) but display similar abnormal accumulation of autophagosomes and lack of acidic autolysosomes as Mon1mut4 mutant cells in starved third instar larvae.

    Expression of Mon1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4hs.PU rescues the loss of acidic autolysosomes (detected by LysoTracker staining) observed in fat cells of starved Mon1mut4/Df(2L)ED7853 third instar larvae.
