Description

A 1.3kb fragment of per 5' flanking region, a short hsp70 activation sequence and a 2kb per 3' UTR region regulate expression of a per cDNA containing a M560D mutation (aimed to destroy a major contact point between Per-Per homodimers), tagged with Tag:MYC at the C-terminus.

In LD conditions, perM560D.T:Hsap\MYC, per01 flies behave similarly to the controls overall, although behavioural anticipation of the D to L and L to D transitions is less pronounced. In DD conditions, only 30-60% of mutant flies are rhythmic and periods are slightly longer than controls, indicating that the circadian clock is impaired in these flies; however temperature compensation is normal.
