A 1.3kb fragment of per 5' flanking region, a short hsp70 activation sequence and a 2kb per 3' UTR region regulate expression of a per cDNA containing W482E and M560D mutations (aimed to destroy major contact points between Per-Per homodimers), tagged with Tag:MYC at the C-terminus.

      perW482E.M560D.T:Hsap\MYC does not restore rhythmic behaviour to per01 flies in LD or DD conditions in more than 20% of flies. The few rhythmic flies exhibit widely variable periods, and temperature compensation is severely compromised.
