Re-integration of DNA into the attP site present in wgKO. The reintegrated sequence is as follows: 1. an FRT cassette containing 'Nrt-wg' (a membrane-tethered, Tag:HA-tagged form of wg in which Tag:M(Nrt) acts as the membrane tethering sequence), 2. wild-type wg cDNA sequence, and 3. a loxP cassette containing a Disc\RFPmCherry.3xP3 marker. The wild-type wg sequence is not expressed due to the presence of the upstream FRT cassette. In addition, a recombined attP/B site is present 5' to the reintegrated sequence.
"wg{KO; NRT-Wg[Body]; WT[Discs]}" larvae expressing wild-type wg in imaginal discs and 'NRT-wg' (a membrane-tethered form of wg) in other tissues have been generated by expressing FLPase in the imaginal discs of homozygous wgKO;FRT.NRT-wg.FRT.wg larvae (converting wgKO;FRT.NRT-wg.FRT.wg to wgKO;-FRT.wg in the discs). These animals largely fail to eclose when co-cultured with control siblings, but eclose into normal-looking adults when grown in separate vials.
