Expression of NetoScer\UAS.cSa in a wild type background under the control of Scer\GAL4G14 results in a reduction in NMJ bouton number and shorter branches. Expression in a mutant background (Neto36) also results in a dose-dependent reduction in bouton number and branching: the higher the expression level of the transgene the more severe the phenotype. Overexpression of Neto also has detrimental effects on the viability of rescued animals at all stages of development. There is a reduction in mEJP frequency compared to controls and a small reduction in amplitude when NetoScer\UAS.cSa is expressed at very high levels. EJP amplitude is also reduced at high expression levels.
Scer\GAL4G14/NetoUAS.cSa rescues Neto36
Expression of NetoScer\UAS.cSa in muscles under the control of Scer\GAL4G14 rescues the embryonic lethality and paralysis seen in Neto36 mutants. Neto36 mutant flies expressing low/moderate levels of Scer\GAL4G14 under the control of Scer\GAL4G14 have significantly reduced spontaneous miniature excitatory junctional potential (mEJP) amplitudes. mEJP frequency is largely normal. Quantal content is significantly increased and evoked EJP is normal.
