Expression of NetoPM.Scer\UAS.T:Avic\GFP under the control of Scer\GAL4G14 is unable to rescue the embryonic lethality and adult viability of Neto36 mutants. Very few animals reach the adult stages and the larvae die during the larval stages. They do not fly and have locomotor defects. The NMJ morphology defects seen in Neto36 are not rescued at moderate levels of expression, and at high expression levels the NMJs are severely altered. The postsynaptic markers are severely disrupted in rescued animals but these post-synaptic defects are not accompanied by cytoskeletal disruption. Rare boutons are seen with no postsynaptic densities (PSDs) and no detectable subsynaptic reticulum (SSR), surrounded instead by dense ribosome fields or myofibrils. The T-bar structures are increased in diameter and are often mis-shaped, collapsed or floating at boutons and there is an increase in synaptic vesicle size. The spontaneous miniature excitatory junctional potential (mEJP) frequency and amplitude are reduced compared to controls. Evoked EJPs are normal. Presynaptic markers are unaffected.
