Description

UAS regulatory sequences drive expression of an Aβ peptide (1-42). The Hsap\APP sequence in the construct is codon-optimized for expression in Drosophila melanogaster. A consensus Drosophila melanogaster translation start site is used. The signal sequence used in the construct is Tag:SS(rPENK).

      Scer\GAL4nSyb.PS>Hsap\APP1-42.Scer\UAS.T:Rnor\SS-PENK flies display greatly reduced median lifespan of only 9 days compared with the average of 30 days for wild-type. Lifespans of the mutants are not significantly different in males when compared to females.

      Flies expressing Scer\GAL4nSyb.PS>Hsap\APP1-42.Scer\UAS.T:Rnor\SS-PENK display extensive locomotor dysfunction.
