Description

UAS drives expression of the entire mRNA sequence of the transcription factor "α1ACT" (with its native start and stop codons), which is one product of the bicistronic Hsap\CACNA1A gene. The CAG repeat/polyQ region that is linked to spinocerebellar ataxia type 6 occurs within α1ACT, and this construct contains a non-pathogenic, wild-type version of 11 repeats. (The normal CAG repeat is between 4 and 18.)

Expressing Hsap\CACNA1AUAS.αACT.Q11 under the control of Scer\GAL4sqh.PW does not lead to defects in lifespan. Scer\GAL4GMR.PU-driven expression does not induce major signs of retinal degeneration.

Expression of Hsap\CACNA1AScer\UAS.αACT.Q11 driven by Scer\GAL4sqh.PW, Scer\GAL4elav.PU, Scer\GAL4Mef2.PU or Scer\GAL4repo.PU does not significantly affect lifespan compared to controls. Expression of Hsap\CACNA1AScer\UAS.αACT.Q11 driven by Scer\GAL4GMR.PU does not cause clear or progressive anomalies in the retina. A more sensitive GFP-based method shows that expression of Hsap\CACNA1AScer\UAS.αACT.Q11 driven by Scer\GAL4GMR.PU is mildly progressively toxic in fly eyes (significant loss at day 7 and 14 but not day 1).
