UASp sequences regulate expression of two copies of the second exon of Torsin cloned in a tail-to-tail configuration separated by a 70 bp spacer, which mediates RNAi against the Torsin transcript.

Expression of TorsindsRNA.Scer\UAS.P\T driven by Scer\GAL4GMR.PF results in a weak rough eye (disorganized retinas) phenotype compared to controls; additional presence of Df(1)bi-DL2 results in a slightly stronger phenotype (though Df(1)bi-DL2/+ alone does not have a rough eye phenotype). At 4 and 7 weeks old, Df(1)bi-DL2/+;Scer\GAL4GMR.PF>TorsindsRNA.Scer\UAS.P\T flies show a widespread significant increase in degenerative spots (holes) on the retina, compared to controls (holes are also significantly increased from 4 to 7 weeks old in mutants). Expression of TorsindsRNA.Scer\UAS.P\T driven by Scer\GAL4GMR.PF results in thickening of pigment granule areas in the retina (38% penetrance) and disorganized pigment granule patterns with occasional gaps or accumulations; remaining retinas do not show a clear thickening phenotype, but show disorganized granule patterns and additional presence of Df(1)bi-DL2 results in stronger phenotypes.
