UAS regulatory sequences drive expression of Hsap\LRRK2 cDNA with a mutation in the GTPase domain (R1441C), fused with an N-terminal Tag:FLAG tag.
Expression of Hsap\LRRK2R1441C.Scer\UAS.cIa.T:Zzzz\FLAG under the control of Scer\GAL4GMR.PU leads rapid age-progressive photoreceptor degeneration in the adult eye leading to visible disorganization of the surface ommatidial array.
Adult flies expressing Hsap\LRRK2R1441C.Scer\UAS.cIa.T:Zzzz\FLAG under the Scer\GAL4Ddc.PL driver are short lived, show a steeper age-related decline of climbing ability and aged flies (6-week-old) display significantly reduced number of dopaminergic neurons from the PPM1/2 cluster compared to controls and a complete loss of PPL1, PAL and PPL2ab neurons, whereas the number of neurons in the PAM and PPM3 clusters is not significantly affected.
