Description

A mutated copy of the Tpi locus (containing the amino acid replacement G74E) has been inserted into the attP site present in TpiattP. This mutation is predicted to result in disruption of dimerization of the Tpi protein.

(FlyBase curator comment: the mutation in the Tpi gene is given as G74E in FBrf0233082, however analysis of the release 6.40 annotated gene model indicates the change to be G75E).

TpiG74E homozygotes are partially lethal; TpiG74E/TpiK11M transheterozygous and TpiG74E homozygous adults, but not TpiG74E heterozygous adults, exhibit an increase in heat sensitivity, shown by the significant decrease in the time until paralysis after a temperature shift to 38[o]C, and an increase in bang sensitivity, shown by a significant increase in recovery time from mechanical stress, as compared to controls; TpiG74E/TpiK11M transheterozygous and TpiG74E homozygous adults also exhibit a significant decrease in lifespan, whereas TpiG74E heterozygous adults exhibit a significant increase in lifespan, as compared to controls.

TpiG74E/TpiG74E-CFP transheterozygotes exhibit an increase in heat sensitivity and an increase in bang sensitivity during adulthood, while showing insignificant changes in the numbers of boutons and branches at the third instar larval neuromuscular junction, as compared to controls.
