A mutated Hsap\TPI1 coding sequence carrying the M82T amino acid replacement is expressed under the control of Tpi regulatory sequences at the endogenous Tpi locus (the human coding sequence replaces the endogenous Drosophila coding sequence). It was inserted into the Drosophila genome via phiC31:int-mediated integration into the attP site present in TpiattP.

One copy of Hsap\TPI1M82T.Tpi (in which the coding sequence of endogenous Tpi is replaced by the human ortologue sequence bearing the M82T amino-acid substitution) in a Tpi-null background (TpihTPI.M82T/Tpisgk-js10) leads to adult exhibiting a small decrease in lifespan, a small increase in bang sensitivity, shown by a small but significant increase in recovery time from mechanical stress, and an increase in heat sensitivity, shown by the significant decrease in the time until paralysis after a temperature shift to 38[o]C, as compared to controls.
