Sequence encoding the internal intrinsically disordered region (IDR) has been deleted from the endogenous Atx2 locus. This deleted region includes a prion-like domain (amino acid residues 420-440) and the endogenous polyQ repeats (amino acid residues 548-656). A GFP tag has been introduced at the C-terminal end of the open reading frame. In addition, a Disc\RFPDsRed.3xP3.cUa marker allele has been inserted into the first intron, transcribed in the opposite orientation to Atx2, as part of the targeting event.
Atx2KI.ΔmIDR-GFP significantly decreases long-term olfactory habituation, but not short-term olfactory habituation, as compared to controls.

At 22[o]C and 25[o]C, expressing Zzzz\poly-GR50.UAS.EGFP under the control of Scer\GAL4GMR.PF induces pupal lethality or visibly necrotic eyes in a considerable proportion of Atx2KI.ΔmIDR-GFP homozygotes; at 18[o]C, only rare death and no eye defects are induced in Atx2KI.ΔmIDR-GFP homozygotes. No defects are induced by Zzzz\poly-GA50.UAS.EGFP expression.
Expressing Hsap\FUSUAS.cBa.Tag:HA under the control of Scer\GAL4GMR.PF induces mild-to-severe eye defects nearly all Atx2KI.ΔmIDR-GFP homozygotes.
