UASt regulates expression of the cDNA LD02314 in which the endogenous signal sequence (position 1-19) is replaced by the signal sequence of CD2 (Tag:SS(rCd2)) and a 10x Tag:MYC epitope, and in which the GPI attachment site is replaced with the transmembrane domain from mCD8 (Tag:M(mCd8a)).

The expression of udtTM.UAS.Tag:MYC,Tag:SS(rCd2),Tag:M(mCd8a) under the control of Scer\GAL4da.G32 rescues both the viability and blood-brain and trachea barrier defects observed in udtf03018 homozygotes. Expression under the control of Scer\GAL4repo only rescues the blood-brain barrier defect, as it does not rescue viability and the trachea is still leaky. Expression under the control of Scer\GAL4en-e16E fails to rescue viability as well as the blood-brain barrier and trachea barrier defects.
