UASt regulatory sequences drive expression of the full-length Tak1 open reading frame (derived from the LD42274 cDNA clone), that has been mutated to carry the K46R amino acid replacement (this is predicted to result in a dominant negative protein). The coding sequence is tagged at the C-terminal end with 3xTag:FLAG.
Third instar larvae expressing Tak1K46R.UAS.Tag:FLAG under the control of Scer\GAL4VGlut-OK371 display a significantly reduced mEPSP (mini excitatory postsynaptic potential) frequency compared to controls. Following high-frequency stimulation, motor neurons display profound short-term facilitation, in contrast to the synaptic depression characteristic of controls. There is no effect on the number of type I boutons and active zones per NMJ, or active zone density.
Motor neurons of third instar larvae expressing Tak1K46R.UAS.Tag:FLAG under the control of either Scer\GAL4VGlut-OK371 or Scer\GAL4C57 display similar mEPSP (mini excitatory postsynaptic potential) and EPSP (excitatory postsynaptic potential) amplitudes and quantal content to controls.
