UASt regulatory sequences drive expression of two open reading frames separated by stop codons (the upstream ORF is simply present to reduce translation of the Tag:Dam-containing downstream ORF). The downstream ORF comprises Tag:Dam, a nuclear localization signal, a Tag:MYC epitope and a tandem dimer of Tag:MCP and is one component of the 'RNA-DamID' system; when expressed together with a lncRNA tagged with Tag:MS2 (binding target of Tag:MCP), it is recruited to sites of lncRNA-chromatin interaction, resulting in methylation of the DNA by Tag:Dam in the vicinity of the interaction site.
