The endogenous RpL22-like locus has been engineered such that the coding sequence can conditionally be removed using FLP recombinase. A pair of FRT sites have been introduced; one is in between the 5'UTR and the start of the coding sequence, the other is downstream of the 3'UTR. A Disc\RFPDsRed(Unk).αTub84B marker has also been introduced downstream of the 3'UTR (but upstream of the second FRT site), so that a conditional knock-out event can be detected in vivo.
Heat shock applied at 1 hour post-egg laying in RpL22-likeCKO animals leads to only two embryos surviving to adulthood while heat shock applied at 24-36 hours post-egg laying leads to a few embryos reaching to adulthood. The rate of survival is less than controls in both situations. However heat shock applied at 48-60 hours post-egg laying in RpL22-likeCKO animals leads to viable adults.
Few RpL22-likeCKO escapers exhibit numerous developmental defects after heat shock applied at 1 hour post-egg laying when compared to controls.
Heat shock at 48-60 hours post-egg laying in RpL22-likeCKO animals leads to fertile females and males without any changes in testis development when compared to controls.
