The RMCE cassette present in TI{TI}rheaC-DsRed has been replaced with sequence that restores the genomic sequence (last two exons of rhea and last exon of CG6638) that was deleted in TI{TI}rheaC-DsRed and also includes a tension-sensing module inserted at the C-terminal end of the rhea protein. The tension-sensing module comprises the YPet and mCherry fluorescent proteins separated by a mechanosensitive linker peptide (HP, Villin headpiece peptide). This transgene acts as as a zero-force control due to the placement of the tension-sensing module at the C-terminus of the rhea protein.
