UASt regulatory sequences drive expression of a mutated form of Lkb1. The C-terminal lipid-binding (LB) motif has been mutated (amino acid substitutions K546A, R547A, R548A, K550A, K551A, K539A, K540A, K541A); this abolishes the ability of the protein to bind liposomes in vitro. In addition, a C564A amino acid substitution (expected to result in a protein that cannot be farnesylated) has also been introduced into the coding sequence. A GFP tag has been inserted at the N-terminal end of the coding sequence.
