20xUAS regulatory sequences (from pJFRC7) drive expression of 'BiPOLES', an optogenetic tool for bidirectional dual-color control of neuron activity. The 'BiPOLES' coding sequence contains both the GtACR2 and Chrimson optogenetic tools, allowing blue-light induced silencing (via the GtACR2 moiety) and red-light induced activation (via the Chrimson moiety) of neuronal cells. The GtACR2 and Chrimson components are joined by a linker composed of a membrane trafficking signal from Kir2.1, a mCer3 fluorescent protein and the transmembrane β helix of the rat gastric H+/K+ ATPase, to maintain the correct membrane topology of both GtACR2 and Chrimson.
