UASt regulatory sequences are fused upstream of the unspliced form of the Xbp1 transcript. This is followed by a Tag:HA tag (in frame with the unspliced form) and then a GFP tag (out of frame with the unspliced form). Under non-stress conditions, the unspliced form of the Xbp1 protein is produced, which is tagged with Tag:HA. Under conditions of ER stress, Ire1-mediated splicing results in a frameshift that brings the GFP tag into frame (and puts the Tag:HA tag out of frame) with the spliced form of the Xbp1 protein.
