The nanos promoter drives expression of 'xCas9(3.7)-ABE(7.10)', a base editor that targets adenine in DNA; it favors the conversion of A to G. The xCas9(3.7)-ABE(7.10) sequence (from Addgene plasmid 108382) encodes a fusion protein composed of 1. a wild-type copy of E. coli tRNA specific adenosine deaminase (tadA), 2. a mutated version of tadA, and 3. x-Cas9(3.7), an engineered Cas9 nickase (contains the D10A mutation) that contains mutations broadening the PAM compatibility to GAA, GAT, and NG in addition to its canonical NGG (see PMID:29512652).
