The nanos promoter drives expression of 'xCas9(3.7)-BE3', a base editor that targets cytosine in DNA; it favors the conversion of C to T. The xCas9(3.7)-BE3 sequence (from Addgene plasmid 108380) encodes a fusion protein composed of 1. a modified rat Apobec1 cytidine deaminase ( RGDID:2133 ), 2. x-Cas9(3.7), an engineered Cas9 nickase (contains the D10A mutation) that contains mutations broadening the PAM compatibility to GAA, GAT, and NG in addition to its canonical NGG, and 3. the uracil DNA glycosylase inhibitor (UGI) from a Bacillus subtilis bacteriophage (see PMID:29512652).
