The RMCE cassettes present in para^MI08578 have been replaced, with the final chromosome likely carrying a single Mi{FlpTag.smV5.0} insertion. The FlpTag conditional tagging cassette in the insertion is in the same orientation as the direction of para transcription. In the absence of FLP, the para gene is tagged with smV5, but in the presence of FLP recombinase, the FlpTag cassette is stably inverted, resulting in loss of this tagging.
