RMCE was used to convert Mi{MIC} insertions to Mi{FlpTag.1} or Mi{FlpTag.2} insertions. In each case, insertions were recovered in each orientation to provide the ability either to include or exclude the GFP tag upon FLP-mediated inversion.
Mi{MIC}ShabMI00848 was converted to Mi{FlpTag.1}ShabMI00848-FT.1-X and Mi{FlpTag.1}ShabMI00848-FT.1.
In this case and the cases below, the insertions written as "-X" are oriented so that the sequence for the GFP tag is not spliced into the mRNA transcript. If the insertion is exposed to FLP recombinase and the FRT-flanked cassette is inverted, the GFP sequence will be spliced into the mRNA transcript. Conversely, the insertions written without "-X" are oriented so that the GFP sequence is spliced into the mRNA. Inversion of the FRT cassette results in the GFP sequence being excluded from mRNAs.
Mi{MIC}ShalMI00446 was converted to Mi{FlpTag.1}ShalMI00446-FT.1-X and Mi{FlpTag.1}ShalMI00446-FT.1.
Mi{MIC}ShMI10885 was converted to Mi{FlpTag.2}ShMI10885-FT.2-X and Mi{FlpTag.2}ShMI10885-FT.2.
Similarly, CRIMIC insertions were converted to TI{FlpTag.1} insertions.
TI{CRIMIC.TG4.1}CG1688CR01570-TG4.1 was converted to TI{FlpTag.1}CG1688CR01570-FT.1-X and TI{FlpTag.1}CG1688CR01570-FT.1.
TI{CRIMIC.TG4.1}eagCR01421-TG4.1 was converted to TI{FlpTag.1}eagCR01421-FT.1-X and TI{FlpTag.1}eagCR01421-FT.1.
A series of FlpTag donor transgenes was created to introduce the smV5 tag into proteins via RMCE in the same way Mi{FlpTag.0}, Mi{FlpTag.1}, Mi{FlpTag.2} and TI{FlpTag.1} transgenes may be used to introduce GFP into proteins. These transgenes are Mi{FlpTag.smV5.0}, Mi{FlpTag.smV5.1}, Mi{FlpTag.smV5.2} and TI{FlpTag.smV5.1}, where 0, 1 and 2 represent different reading frames.
Mi{MIC}IhMI12136 was converted to Mi{FlpTag.smV5.2}IhMI12136-FT.smV5.2-X and Mi{FlpTag.smV5.2}IhMI12136-FT.smV5.2.
Mi{MIC}ShMI10885 was converted to Mi{FlpTag.smV5.2}ShMI10885-FT.smV5.2-X and Mi{FlpTag.smV5.2}ShMI10885-FT.smV5.2.
Mi{MIC}ShabMI00848 was converted to Mi{FlpTag.smV5.1}ShabMI00848-FT.smV5.1-X and Mi{FlpTag.smV5.1}ShabMI00848-FT.smV5.1.
TI{CRIMIC.TG4.1}eagCR01421-TG4.1 was converted to TI{FlpTag.smV5.1}eagCR01421-FT.smV5.1-X and TI{FlpTag.smV5.1}eagCR01421-FT.smV5.1.
TI{CRIMIC.TG4.1}CG1688CR01570-TG4.1 was converted to TI{FlpTag.smV5.1}CG1688CR01570-FT.smV5.1-X and TI{FlpTag.smV5.1}CG1688CR01570-FT.smV5.1.
Mi{MIC}ShalMI00446 was converted to Mi{FlpTag.smV5.1}ShalMI00446-FT.smV5.1-X and Mi{FlpTag.smV5.1}ShalMI00446-FT.smV5.1.
Mi{MIC}ShawMI01735 was converted to Mi{FlpTag.smV5.1}ShawMI01735-FT.smV5.1-X and Mi{FlpTag.smV5.1}ShawMI01735-FT.smV5.1.
Finally, Mi{FlpTag.smV5.0}paraMI08578-FT.smV5.0-X and Mi{FlpTag.smV5.0}paraMI08578-FT.smV5.0 were generated from a progenitor stock with two side-by-side Mi{MIC} insertions in the para gene (Mi{MIC}MI08578a Mi{MIC}MI08578b) with 37 bp of unknown origin between them. It is likely that RMCE resulted in the final chromosome carrying a single Mi{FlpTag.smV5.0} insertion composed of the left end of one progenitor Mi{MiC} and the right end of the other progenitor Mi{MIC} with intervening sequences replaced by Mi{FlpTag.smV5.0} sequences, but it is possible that RMCE swapped the internal contents of both insertions so that two Mi{FlpTag.smV5.0} insertions are now present.
