Description

Sequence encoding a Gly-Ser linker, mCherry and pHluorin(Unk) has been inserted into the endogenous stan locus at the C-terminal end of the coding sequence of the stan-RA isoform. The mCherry-pHluorin(Unk) dual fluorophore can act as an acidification sensor in vivo as pHluorin(Unk) is pH sensitive but mCherry is pH-resistant, meaning that stan protein that is present in degradative compartments (red fluorescence only, from mCherry) can be distinguished from potentially active stan protein in other compartments, including on the cell surface (yellow fluorescence due to the combination of mCherry and pHluorin(Unk) fluorescence).
