Description
Lipases, generally defined as triacylgycerol hydrolases (EC 3.1.1.3), cleave the carboxylester linkages in triacylglycerol (TAG) to produce diacylglycerol (DAG) and a carboxylate. In insects, TAGs are used for energy storage and for storage of fatty acid precursors for semiochemicals, while DAGs are the main form of transported lipid. Lipases thus control key steps in lipid uptake, transport and utilisation. The acid lipases or abhydrolipase family are so named because the founding member, the gastric lipase of mammals, was found to function at acidic pH. The acid lipases predominantly hydrolyse TAGs and cholesterol esters, and are defined by Pfam domain PF04083 (corresponding to InterPro IPR006693). (Adapted from FBrf0208359.)
Notes on Group

Bacterially expressed Lip3 has weak TAG lipase activity on short-chain fatty acid glyceryl trioctanoate but no detectable activity on glyceryl trioleate, a more physiological substrate (FBrf0243470). Lip3 expressed in mammalian cell culture does not have TAG, DAG or MAG activity - instead, lipidomics analysis of a Lip3 mutant suggests Lip3 is a putative phospholipase (FBrf0254005).
