Glutathione (GSH) catabolism occurs predominantly at the cell surface where GSH is cleaved into L-cysteinylglycine and an L-glutamyl amino acid. L-cysteinylglycine can be broken down at the cell surface by dipeptidases into L-cysteine and glycine followed by their import into the cytosol. Alternatively, L-cysteinylglycine is imported into the cytosol prior to its cleavage into L-cysteine and glycine. The L-glutamyl amino acid is also imported into the cytosol where it is converted into L-aspartate. Cytosolic GSH can also be catabolised in the cytosol; however, this is likely to be a minor pathway. L-cysteine, glycine and L-aspartate are recycled back to synthesize GSH.
