Candidate cell surface proteins were screened for binding to an alkaline phosphatase tagged construct encoding the Ptp10D extracellular domain. 976 candidate genes encoding predicted cell surface proteins were identified, of which 311 genes had stocks available with upstream UAS insertions. Each of these 311 genes was ectopically expressed in pan-muscle or pan-neuronal patterns using the GAL4-UAS system, and live embryos were dissected and incubated with AP-tagged Ptp10D.
Source was live embryos of transgenic fly line; bait produced from tagged transgenic construct; prey derived from baculovirus-infected insect cells.
Interaction in vitro; bait produced as a recombinant fusion protein in baculovirus-infected insect cells; prey produced as a recombinant fusion protein in baculovirus-infected insect cells.
