Insertion lines from this collection were assessed for inclusion in the Gene Disruption Project collection.
A genetic screen allowing selection new insertions on the autosomes was carried out. Mobilization of the P{lacW} element occurred in males carrying a w[-] allele and P{lacW} on the X chromosome, and P{Δ2-3}99B, a stable source of P transposase, on a marked third chromosome. A singe w[+] F1 male from each dysgenic cross was used for subsequent characterization, to avoid pre-meiotic clusters. Lethal insertions were selected and balanced stocks created. 2355 second chromosome insertion lines and 160 third chromosome insertion lines were retained for further characterization.
Markers in stocks (as originally submitted to the BDSC):w*.
Flanking genomic sequences were obtained by sequencing of inverse PCR products, and were mapped by alignment to the genomic sequence. A final set of 854 independent insertion lines was selected for extensive characterization, representing 783 genes on II and 67 genes on III.
