A set of transgenic insertion stocks derived by TE mobilization using the Tni\piggyBac-based protein-trap construct PBac{HpaI-GFP.A}. The construct carries a y+mDint2 mini-yellow visible marker and an Avic\GFP vital fluorescent protein-trap marker.
Use of a high-throughput embryo sorter allowed screening of very large numbers of animals for GFP-expressing protein trap insertions.
Mobilization of the Pbac construct occurred in animals carrying PBac{HpaI-GFP.A} on a second chromosome CyO balancer chromosome and a source of Tni\piggyBac\T transposase on the third chromosome. Avic\GFP expression was screened in embryos using a COPAS Drosophila embryo sorter (Union Biometrica).
Flanking genomic sequences were obtained by sequencing of inverse PCR products, and were mapped by alignment to the genomic sequence.
