This collection is based on cDNAs from the BDGP Gold Collection (https://www.fruitfly.org/EST/gold_collection.shtml)
cloned into pDNR-Dual from Clonetech. The Open (O) collection lacks the native stop codon and is appropriate for creation of carboxy-terminal fusion proteins. For expression, transfer of ORFs is mediated by in-vitro Cre-lox recombination to appropriate expression vectors, using terminal tags of choice. [https://www.fruitfly.org/EST/proteomics.shtml]
Several widely used Drosophila expression vectors have been converted, via insertion of a single loxP site, to accept ORFs from the ExpressR-NS and ExpressR-O clones to generate both 5' and 3' fusions. These vectors are used for inducible protein expression in tissue culture cells under the metallothionein-inducible promoter (pMK33-based vectors) and in transgenic flies under GAL4-inducible UAS promoter (pUAST-based vectors). A list of available clones can be downloaded from the DGRC: https://dgrc.bio.indiana.edu/include/file/DGRC-ORF-clone-list-2021-02-10.xlsx; clones from this collection are designated 'BO'.
