The pBS-KS-attB1-2-PT-SA-SD-1-HRP-S plasmid contains a recombination-mediated cassette exchange (RMCE) cassette that consists of two inverted attB sites which surround a protein trap sequence composed of a splice acceptor site, a (GGS)[[4]] flexible linker, a multiple tag cassette (HRP-Tag:S), another (GGS)[[4]] flexible linker and a splice donor site. In the presence of phiC31:int integrase, the attB cassette can undergo RMCE with a target in the genome that contains inverted attP sites, such as the Mi{MIC} element, replacing the target attP cassette with the donor attB cassette sequence. Integration into a Mi{MIC} insertion in a coding intron of a native Drosophila gene of interest in the correct orientation and intron phase converts the insertion into a protein trap, tagging the Drosophila protein with the multiple tag sequence (HRP-Tag:S). The pBS-KS-attB1-2-PT-SA-SD-1-HRP-S plasmid is designed for integration into a phase 1 coding intron.
