Description

Cloning vector that contains a single sgRNA scaffold into which two Tag:BS(MS2-CP) stem loops have been inserted (one on the tetraloop and one on stem loop 2) and which is expressed under the control of the constitutive ' U6:2 ' (snRNA:U6:96Ab) promoter. An sgRNA that targets sequence upstream of a gene of interest can be cloned into the vector, and the attB site present can be used to integrate the sequence into an attP-containing docking site in the genome. The resulting transgenic construct can be used as one component of the three-component flySAM (synergistic activation mediator) system, to produce effective transcriptional activation of the gene of interest.
