The pCFD8 vector is designed to facilitate cloning of one or multiple LbCas12a-specific crRNAs downstream of the strong, ubiquitous U6:3 (snRNA:U6:96Ac) promoter. No flanking sequences are required in between crRNAs in an array, as LbCas12a has the ability to directly process arrays into individual crRNAs. A downstream tRNA is present in pCFD8, as it was shown previously (FBrf0233589) that a flanking tRNA can enhance gene editing by Cas12a type crRNAs, but this requirement has not been assessed for constructs derived from pCFD8. The vector contains an attB site, allowing integration into an attP-containing docking site in the genome to produce a transgenic construct.
