The pCFD5-NS dual-expression plasmid is designed for expression of a prime editing guide RNA (pegRNA) plus a nicking sgRNA for use in the 'Prime editor 3' (PE3) system. The pegRNA and sgRNA are expressed under the control of the strong, ubiquitous U6:3 (snRNA:U6:96Ac) promoter as a single polycistronic tRNA-based cassette, and the separate pegRNA and sgRNA components are then released by the endogenous tRNA-processing machinery. The vector contains an attB site, allowing integration into an attP-containing docking site in the genome to produce a transgenic construct.
