Cloning vector used to generate transgenic constructs in which a gene of interest can be expressed, singly or in combinations, via three binary expression systems. The vector contains an expression cassette composed of an mFRT71 cassette containing 5xlexAop regulatory sequences plus a minimal Hsp70 promoter ('Module 1'), an FRT cassette containing 5xUAS regulatory sequences plus a minimal Hsp70 promoter ('Module 2'), a loxP cassette containing 5xQUAS regulatory sequences plus a minimal Hsp70 promoter ('Module 3'), a multiple cloning site, and a ftz intron followed by the SV40 polyA signal (to ensure transcription termination and mRNA stability). An attB site is present downstream of the expression cassette (to facilitate generation of transgenic animals using phiC31:int-mediated integration. The vector is marked with w+mC and both the expression cassette, attB site and marker are flanked by P-element ends. Each of the three regulatory sequence modules is flanked by incompatible recombination sites, allowing them to be excised singly or in combination depending on the specific recombinase(s) used.
