Subject: FlyBase query
Dear Dr. Luo,
I am curating your DIS article for FlyBase:
Luo et al., 1999, D. I. S. 82: 102--105
In this you describe an enhancer trap vector PFLP. I am writing to
ask for some more details about this construct, such as the marker
allele in the vector and the promoter that drives the FLP gene. This
information would be recorded in FlyBase as a 'personal communication to FlyBase
from you to FlyBase.
You state that you 'inserted the open reading frame of the yeast FLP
recombinase into an enhancer trap vector' which comes from:
Giniger et al., 1993, Roux Arch. dev. Biol. 202: 112--122
The enhancer trap vector in that paper is 'P{KZ.TRAP}', which contains
a kinesin-lacZ fusion gene expressed under the control of a P-element
transposase promoter.
Did you just replace the kinesin-lacZ fusion gene with FLP sequences to
make your PFLP vector ? - then I will know that the promoter driving
the FLP coding region is the P-element transposase promoter, and also
that the marker gene is w, since this is the marker gene in the
P{KZ.TRAP} vector,
I look forward to hearing from you,
Gillian

Dear Dr. Millburn:

Yes we did insert FLP to replace KZ without changing anything else to
the vector.

That's correct.
Best wishes,
