I noticed that the single flanking sequence (GenBank CZ481614) has two segments that align on opposite strands 263 kb apart. The sequence is high quality and the alignments are unambiguous. A segment of 43 bp, which abuts the piggyBac 5prime end (positions 130-172 of the GenBank sequence), maps to X-3C2 release 5 position 2751110. The other segment (positions 3-130 of the GenBank sequence) maps to X-3C7 release 5 position 3014490 in the kirre gene. I agree with you that the sequence accession looks good. My initial inclinations: 1) The location of the insertion itself should be revised to 2751109. 2) The kirree03862 allele should remain, but be dissociated from the insertion. 3) Your comments should show up in the reports for both (maybe with some sort of "Caution" flag). --Lynn
