Determination of cell line sex by analysis of roX1 and roX2 titers.

I have determined the sex of several cell lines by analysis of roX1 and roX2 titers. This was done by comparing each cell line to Kc167 (FBtc0000001) using DGRC-2 transcriptome microarrays. The parental line Kc (FBtc0000998 ) is known to be female, as judged by the dsx splicing pattern (FBrf0089731), and I found no detectable expression of roX1 or roX2 in Kc167 on my microarrays. The following cell line was deemed to be female, because it also had no detectable signal from either roX1 or roX2: ML-DmD16-c3 (FBtc0000097). The following lines were deemed to be male, because they had strong expression from both roX1 and roX2: ML-DmBG2-c2 (FBtc0000053), ML-DmD21 (FBtc0000086), ML-DmD20-c2 (FBtc0000109), ML-DmD20-c5 (FBtc0000112), and CME W1 cl.8+ (FBtc0000151).
