I'd like to submit corrected coordinates for two Exelixis insertions. Bob Levis looked back in Gene Disruption Project files and supplied the corrected coordinates.
FlyBase currently lists the Release 5 breakpoint of PBac{WH}pdm3f00828 as 2R:4732831 within the sns gene in polytene region 44F. The correct coordinate is 2R:4281766 (R5) within the pdm3 gene in 44C. This is consistent with the alignment of flanking sequences CZ483312 and CZ483313.
One consequence of the corrected coordinate is that the breakpoints of Df(2R)BSC267 change from those currently listed in FlyBase. Rather than 2R:3970399 ;4732831 (R5) with estimated cytological breakpoints 44A4;44F1, the coordinates should be 2R:3970399 ;4281766 with estimated breakpoints 44A4;44C4.
FlyBase currently lists the Release 5 breakpoints of PBac{WH}f04065 as X:16629054 within the TwdlY gene in 15A. The correct coordinate is X:16526135 (R5) within the CG9777 and rok genes. This is consistent with the alignment of flanking sequence CZ486631. The changed coordinate will require the elimination of the TwdlYf04065 allele (FBal0218022) and the creation of rokf04065 and CG9777f04065 alleles.
There are two consequences of the corrected coordinate. The ends of Df(1)BSC760 change from X:16420365 ;16629054 (R5) with estimated cytological breakpoints 14E1;15A3 to X:16420365 ;16526135 (R5) with estimated breakpoints 14E1;14F2. The ends of Df(1)FDD-0331226 change from
X:16629054 ;16730272 (R5) with estimated breakpoints 15A3;15A11 to X:16526135 ;16730272 (R5) with estimated breakpoints 14F2;15A11. These corrected breakpoints are consistent with the Minute phenotype of Df(1)FDD-0331226 heterozygotes from deletion RpS19a and the lack of a Minute phenotype for Df(1)BSC760 heterozygotes.
Thanks!
Kevin
