The following information accompanied stocks donated to the Bloomington Stock Center by Kent Nybakken, Boston Biomedical Research Institute.
To produce P{UAS-deltaCOP.HA}, the full length coding sequence from deltaCop (FBgn0028969), lacking only the C-terminal stop codon, was PCR'ed using primers engineered to produce a 5' in-frame Bam HI site and an in-frame 3' Not I site. This fragment was digested with Bam HI and Not I and ligated into a vector with a 3' triple hemagglutinin tag. The C-terminal triple HA tagged form of deltaCOP was excised by cleavage with Bam HI and Xba I and ligated into Bgl II-Xba I cut pP{UAST}.
P{UAS-deltaCOP.HA}2M is a homozygous viable and fertile, second chromosome insertion.
P{UAS-deltaCOP.HA}5M is a third chromosome insertion.
