To generate P{UAS-CkIalpha.HA}, the Casein kinase Ialpha (FBgn0015024) cDNA LD05574, obtained from the Drosophila Gene Collection, was used as a template to PCR the full-length codons of CkIalpha minus the stop codon and simultaneously add a 5' Bam HI site and a 3' Not I site. This PCR fragment was digested with Bam HI and Not I and ligated into a vector made by Kent Nybakken containing an in-frame, C-terminal, triple hemagglutinin (HA) tag followed by a stop codon. The full-length, tagged cDNA was then excised using Bam HI and Xba I and inserted into Bgl II-Xba I cut pP{UAST}.
P{UAS-CkIalpha.HA}1 is a second chromosome insertion.
P{UAS-CkIalpha.HA}5 is a third chromosome insertion.
