CG5280T317 was induced with EMS on a chromosome carrying the homozygous viable and fertile P{EPgy2}mldEY12290 insertion.
CG5280T317 primarily affects the speed or efficiency of sperm entrance into seminal receptacles of females. In matings with wild-type males, seminal receptacles were fully filled 20 minutes after mating, whereas only a few sperm enter the seminal receptacles 20 minutes after mating with CG5280T317 homozygous males.
CG5280T317 was mapped by meiotic recombination to 61F and its position was narrowed to 3L:8972087..8991989 (R5) in crosses to deletions. It failed to complement Df(3L)ED4421, Df(3L)ED4416, Df(3L)Scf-R11, Df(3L)Scf-R6 and Df(3L)BSC130, but complemented Df(3L)DocA, Df(3L)ED4414 and Df(3L)ED4413. Of the four annotated genes in this DNA interval, CG5280 is the only gene expressed in the testis. The SnpEff program (FBrf0217795) identified a premature termination at the 39th codon (W39*) in CG5280. No mutations were found in the three remaining genes in the region. Consequently, CG5280 is likely the gene responsible for the sperm storage defect, though this has not been confirmed by regular DNA sequencing or Western analysis.
