GalphaoA221D - CRISPR/Cas9-mediated homologous dependent repair was used to put amino acid change A221D into Galphao. This change mimics the A221D change found in a human GNAO1 variant associated with spastic paraparesis and dystonia. The A221D change was put in along with a 3xP3-DsRed marker flanked by PBac ends that was then removed via PBac transposition. Transposition leaves behind a TTAA in an intron.
GalphaoG45E - CRISPR/Cas9-mediated homologous dependent repair was used to put amino acid change G45E into Galphao. This change mimics the G45E change found in a human GNAO1 variant associated with epilepsy-related symptoms. The G45E change was put in along with a 3xP3-DsRed marker flanked by PBac ends that was then removed via PBac transposition. Transposition leaves behind a TTAA in an intron.
TI{TI}GalphaoKO - CRISPR/Cas9-mediated homologous dependent repair was used to replace a 76 bp region just before the 2nd coding exon of Galphao with a cassette carrying stop codons and lox-P flanked 3xP3-RFP. This results in a null allele of Galphao.
