The sequence information for AGO21-1-7, AGO22-5-14, and AGO22-16-4 is in the associated file. Below are some notes on the creation of the lines.
    Three AGO2 null mutants were obtained, AGO21-1-7, AGO22-5-14 and AGO22-16-4. The genotypes are y2w; AGO2*/TM6B,Hu,Tb.
    AGO21-1-7 has a 7bp deletion in the coding region of AGO2. There is no AGO2 expression due to the accompanying deletion in the AGO2 promoter region.
    RNAi activity was checked using a GMR-wIR fly, and these mutants have lost RNAi activity.
    Protein expression data is in the associated file. The 4D2 antibody from Siomi's lab was used.
    The AGO2 mRNA expression level was shown to be very low in the 3 mutants.
    The three null mutants are all homozygous viable.
    The AGO21-1-7 homozygous mutant line is very healthy and can be kept as a stock.
    The homozygous AGO22-5-14 and AGO22-16-4 lines are unhealthy and cannot be maintained as healthy stocks.
    It is not clear why the phenotypes are different between the mutants; the background may be a bit different.
