P{UAS-Doa.91kD} expresses the 91 kD Doa PK isoform under UAS control. The construct was generated using pP{UAST} and cDNA clone LD31161. This cDNA includes sequences introduced by alternative splicing that encode six amino acids (NHTLYS) in the Doa catalytic domain eight amino acids downstream of the LAMMERILG sequence. The sequences encoding these six amino acids are present in Doa genes from other insect species, but, as of this writing (June 2023), evidence for the presence of these amino acids in proteins has been seen in Drosophila melanogaster only in the 91 kD Doa isoform.
P{UAS-Doa.91kD}1 is a second chromosome insertion.
P{UAS-Doa.91kD}3 is a third chromosome insertion.
The expression of both insertions under the control of a GMR-GAL4 driver results in slightly rough eyes. These phenotypes are less severe than those resulting from ectopic expression of the Doa 55 kD or 69 kD isoforms with the same driver.
