The following information accompanied stocks donated to the Bloomington Stock Center by Lenny Rabinow, Harvard Medical School. Doa2C is a six base-pair deletion of a Met codon and the adjacent Pro codon (residues 12 and 13) of the RJ, RN, RT and RU transcripts encoding the 69 kD Doa isoform expressed only in female bodies (but in the heads of both sexes). The relevant wild type sequence is: TGCAAACACTCGCACCTAAGCAGAAACAACAGTACAAAACCCATGCAACTACCCTCGCTGAAGGACAAGCTCATGCCCAGCGGCAGCGTGCAGCAAGCGAAAGCGAATTTCTCCTGGCAATCGCTAACGCAACTCTTGAGCGGCCTGTGG encoding 1 MQLPSLKDKLMPSGSVQQAKANFSWQSLTQLLSGLWQRLYLPRSPFLAL 49 The Doa2C sequence is: TGCAAACAGNTNGCACCTAAGCAGAAACAACAGTACAAAACCCATGCAACTACCCTCGCTGAAGGACAAACTCA------GCGGCAGCGTGCAGCAAGCGAAAGCGAATTTCTCCTGGCAATCGCTAACGCAACTCTTGAGCGGCCTGTGG encoding MQLPSLKDKL-SGSVQQAKANFSWQSLTQLLSGLWQRLYLPRSPFLAL Doa2C does not suppress wa phenotypes as seen with mutations affecting other Doa isoforms. Homozygotes show reduced viability and fertility. Progeny of homozygous females have frequent embryonic cuticular defects, which are not rescued by being fertilized by wild type males.
