P{jbug.GFP} consists of a genomic fragment that encompasses the entire jbug transcription unit, with the sequence for a GFP tag inserted into the sixth coding exon 6 ( 2R:22731144-22731492 (R6)). It results in the expression of GFP-tagged proteins from jbug transcripts RF, RL and RN and the expression of untagged proteins from all other transcripts under native control. It can rescue both the lethality and wing blister phenotypes of alleles of the moa complementation group.
P{jbug.GFP.MGGM} consists of a jbug genomic fragment truncated after the exon containing the first transcriptional stop. It contains the same GFP tag sequence as P{jbug.GFP}. It results in expression of GFP-tagged proteins from jbug transcript RL and the expression of untagged proteins from transcripts RC, RG, RH, RJ and RK under native control. It can rescue the wing blister phenotypes of alleles of the moa complementation group.
P{jbug.GFP.MGcDs} consists of a jbug genomic fragment lacking the second transcriptional start site (deletion of 2L:22725543-22727564 ;22729274-22729816 (R6)). It contains the same GFP tag sequence as P{jbug.GFP}. It results in expression of GFP-tagged jbug proteins from transcripts RF, RL and RN and the expression of untagged protein from transcript RM under native control. It can rescue the wing blister phenotypes of alleles of the moa complementation group.
In addition to the rescue data, mutations have been found within the jbug transcription unit in alleles of the moa complementation group. Together, this evidence indicates that alleles of the moa complementation group are mutations of the jbug gene.
P{jbug.GFP}191c, P{jbug.GFP.MGGM}M23.1 and P{jbug.GFP.MGcDs}M29 are third chromosome insertions.
P{UAS-jbug.T} was generated by Xiao Tan. It expresses the jbug RL transcript encoded by cDNA clone RE40504 under UAS control.
P{UAS-jbug.T}M5 is a third chromosome insertion.
